STAINING EFFICACY OF ALTA IN COMPARISON WITH EOSIN STAIN: A HISTOLOGICAL STUDY ON ORAL TISSUES

Background: Histological staining is a series of processes undertaken for tissue staining procedures. In routine use, H&E is used for tissue staining. Alta is a cosmetic dye manufacture. The stain is used by women to adorn their hands and feet during function in many parts of India. It imparts a bright red colour on the skin. The constituent of the stain is Crocein scarlet and Rhodamine B. The aim of the study is to assess the staining proficiency of Alta on various oral histological tissues and compare the staining efficiency with that of Eosin. Materials & Methods: Various tissues such as skin, lymph nodes, mucosa, salivary gland, muscles and mucosa were selected for tissue sections. Sectioning of the tissues was done in two batches. One batch was stained with Alta and the other batch was stained with H&E. Assessment was done by a single blinded observer. Scores were tabulated and statistical analysis was made using SPSS version 20. Results: Alta has given better results in nuclear and cytoplasmic intensity and cytoplasmic and nuclear morphology. Mann-Whitney U tests p value > 0.05, hence the association is statistically not significant. Conclusion: Alta is more efficient than Eosin in staining histological slides.
